Countable Labs, the creator of Countable PCR, a breakthrough single-molecule PCR technology that is redefining the way science measures biology, announced a collaboration agreement with Promega, a leader in providing innovative solutions and technical support to the life sciences industry.
The deal brings together Promega Maxwell’s capabilities® Nucleic acid extraction system and quantitative PCR instruments for complete, unbiased DNA and RNA extraction and quantification, combining products and expertise to offer customers a complete, practical and easy-to-use solution for DNA and RNA quantification.
The combined workflow leverages Promega Maxwell® System, a reliable, fast and simple automated bench-top nucleic acid extraction instrument that uses a particle-based paramagnetic workflow to process up to 48 samples in parallel, providing high-quality nucleic acids for a wide range of downstream applications. Nucleic acids are then analyzed with quantitative PCR instruments, isolating individual molecules in 30 million compartments for true single-molecule quantification and using the majority of the extracted material as input – probing the samples more fully to maximize the detection of rare targets that other methods miss.
In a poster published by Countable Labs illustrating the utility of the workflow, total cfDNA was extracted from human blood plasma using an automated magnetic bead-based system (the Rapid ccfDNA kit at Maxwell® CSC system) to ensure consistent recovery from low inflow material. When combined with a pre-amplification step, a multiplexed KRAS assay using measurable PCR reached 0.08% MAF for G12C without compromising sensitivity for multiplexing. Results were reproducible in technical replicates and achieved with minimal optimization.
