NIH -funded study uses peak imaging techniques to reconstruct features based on learning and memory in the mouse brain

What:

In a study supported by National Institutes of Health (NIH), researchers revealed the structural foundations of memory formation in a wide network of neurons in the mouse brain. This work sheds light on the fundamentally flexible nature of the way in which memories are made, describing the changes related to learning in cellular and sub -cells with unprecedented analysis. Understanding this flexibility can help explain why memory and learning processes sometimes go wrong.

The findings published in the ScienceHe showed that the neurons assigned to a trace of memory reorganized their connections to other neurons through an informal type of connection called multi-conventional bouton. In a multi-conventional bouton, the axis of the neuron that relays the signal with information of multiple neurons that receive the signal. According to the researchers, multi-conventional boutiques can allow the cellular flexibility of coding information observed in previous research.

The researchers also found that the neurons involved in memory formation were not preferably associated with each other. This finding disputes the idea that “the neurons that trigger together flock together”, as will be predicted by a traditional theory of learning.

In addition, the researchers noticed that the neurons that were allocated to a trace of memory reorganized some intracellular structures that provide energy and support and plasticity to neuronal connections. These neurons also had enhanced interactions with support cells known as astrocytes.

Using a combination of advanced genetic tools, 3D electron microscopy and artificial intelligence, scientists at Scripps Marco Uytiepo, Anton Maximov, Ph.D. And their partners were rebuilt a wiring diagram of neurons involved in learning and identified structural changes in these neurons and their connections to cellular and sub -cell levels.

To examine the structural characteristics associated with learning, the researchers exposed mice into an air conditioning task and examined the hippocampus area about 1 week later. They were chosen this point in time because it appears after the first coding of memories, but before reorganized for long -term storage. Using advanced genetic techniques, researchers who are permanently highlighted by Hippocambu neurons activated during learning, which allowed reliable identification. They then used 3D electronic microscopy and artificial intelligence to produce nanocycin rebuilding of stimulant neural networks involved in learning.

This study provides a complete picture of the structural characteristics of memory formation in an area of the brain. It also raises new questions for further exploration. Future studies will be vital to determine whether such mechanisms operate at different times and nerve circuits. In addition, further exploration of the molecular composition of multiple synchronic boutiques to determine their exact role in memory and other cognitive processes.

The research was supported by funding by the National Institute of Mental Health, the National Institute of Neurological Disorders and Stroke and NIH Brain research through promotion of innovative neurotechnologies® Initiative or Brain Initiative®.
