Breast cancer is one of the most common cancers among women in the United States. Thanks to decades of fundamental research, it is also one of the most therapeutic. The exception is a particularly aggressive variant known as a triple negative breast cancer (TNBC). It is 10 to 15 percent of all cases of breast cancer. It disproportionately treats younger and African American women. There are no effective treatments. A new discovery by Cold Spring Harbour David Spector and Postgraduate Student Wenbo Xu-Publed at Molecular CancerA American Union magazine for cancer research-could help the first.

The Spector team researchs the role of RNA called long -coded RNAS (LNCRNAS) in the formation of cancer. They have now discovered that a rarely studied LNCRNA, previously involved in gastric cancer, also plays part in TNBC. Is called LINC01235. Found the Spector Laboratory LINC01235 When performing RNA sequence in human breast organism tumor microscopically three -dimensional cancer models. Xu, who pioneered the project, suspected a connection between LINC01235 and TNBC when reviewing data by Atlas of cancer genome, a database of more than 11,000 cancer patients. Data suggested expression of LINC01235 is associated with that of breast cancer gene Nfibalready connected to TNBC.

Still, “very little” were known Nfib And next to nothing LINC01235SPECTOR says. So the team decided to “sink deep into it”. They had important questions: “How does it work in the cell? What processes could be involved?

To test the XU case, the team hit out LINC01235 in cancer cells, using CRISPR. Separately, they were hit in cancer cells and organoids, using anti -amidal molecules. In both experiments, they found that RNA’s reduction also declined also Nfib expression. When this happened, the formation of TNBC organizations is suppressed.

Our findings prove that LINC01235 It regulates positively Nfib transcription. This regulates Notch Street, affecting the proliferation of cells in the progress of TNBC. ”

Wenbo Xu, postgraduate student

More research is needed for scientists to find a possible treatment for TNBC. However, the Spector says, these findings emphasize the importance of non -coded RNA in this search: “The goal here is to understand the mechanisms by which cellular functions and the way diseases undertake these functions, perhaps by increasing the RNA molecule or regulation of a RNA molecule.

Everyone is another link in the chain. So, LINC01235 Offers a crucial step in the right direction.
