Dental caries or tooth decay is a common oral health that often causes significant pain and discomfort and can even lead to tooth loss. In severe and unprocessed cases, bacterial infection combined with host immune response can cause bone absorption or bone tissue distribution at the root of the teeth. In addition, traditional treatments for advanced dental caries, such as surgery, can lead to bone defects that require complex bone vaccination processes.

Based on this knowledge, bone mechanics and regeneration of dental tissues have gained the attention of researchers worldwide. Recent reports indicate that the micronas (Mirnas) -mall, non -coding ribonucleic acid sequences play a key role in bone regeneration. However, the underlying mechanisms and trails regulated by Mirnas remain unclear.

To investigate the inherent procedures involved in bone dental repair, a team of researchers led by Associate Professor Nobuyuki Kawashima, a postgraduate student Ziniu Yu and Professor Takashi Okiji from the Postgraduate School Their findings were published in Volume 23 and the Issue 189 of Newspaper of translation medicine On February 16, 2025.

“HDPSCS is a type of mesenchymal stem cell that are capable of differentiating either in toothplanets or in osteoblasts, key players in dental repair,” Kawashima explains. “In our study, we focused on a molecule called Mirna-27A, which we have found that we have an anti-inflammatory effect with the suppression of NF-KB Street, but can also promote tissue regeneration, activating WNT and BMP signaling.

Initially, scientists used tools based on bioinformatics to explore the effects of MIRNA-27A over-expression on HDPSCS. They identified protein 3 (SOSTDC1) associated with Dickkopf (DKK3) and Protein 1 (SOSTDC1) as the main target genes regulated by Mirna-27A. In addition to DKK3 and SOSTDC1, other negative regulators of the family signaling (WNT) of the family (WNT) playing a key role in shaping new bone and dental tissues. In addition, including the axis 2 inhibition protein and the adenomatous covered coli, they were also adjusted down to HDPSC over-expressed MIRNA-27A. This suggests that the Mirna-27A helps to raise these organic brakes, allowing cells to more effectively activate bone formation signals.

In addition to the stimulation of WNT Street, the Mirna -27A was found to significantly affect the Dento/Osteoblastic differentiation of HDPSCs and to activate the bone protein pathway (BMP). The activation of both WNT and BMP streets suggests that hard tissue formation cells were promoted by differentiation of HDPSCs.

To validate their findings, researchers transplant collagencons containing HDPSCs expressing Mirna-27-27- express to artificial defects that were introduced into the mice’s bone bone. Subsequent analyzes revealed the formation of a new bone tissue, which was absent in the control group.

Kawashima concludes by highlighting the therapeutic potential of research: “These results suggest that the Mirna-27A could play a central role in encouraging bone-like tissue formation.

In summary, this study underlines the important translation potential of Mirna-27A in promoting dental tissue regeneration.
